It was a new personal best (PB) for Banjamin, whose previous PB of 36:04 was set at the 2013 U Run.
18,000 runners took part in the Nike We Run Singapore that was flagged off at the F1 Pit Building.
A unique 10km route with various split points will allow runners to choose different terrain, conditions and atmosphere.
The spotlight was on Singaporean veteran, Vivan Tang, 43, who hit the finish tape in 40 minutes 11 seconds to win the women's 10km race at the Yellow Ribbon Run.
Singaporean Wong Hoong Wei, 33, was the fastest finisher at the Sundown 100km Ultra Marathon.
Mok managed to shave 6 minutes 43 seconds from his 2012 timing where he finished third behind then winner Soh Rui Yong and runner-up Ashley Liew, both of whom did not take part this year.
Mok Ying Rong, 19, defended her 15km title at the 2013 Race Against Cancer, convincingly finishing in 1 hour 6 minutes and 13 seconds. It was an improvement of 31 seconds over her timing last year.
